As incretinas são hormonas libertadas pelo intestino em resposta à ingestão de nutrientes que potencializam a secreção de insulina estimulada pela glicose. A pesquisa sobre as incretinas foi motivada pela observação de que a administração de uma carga oral de glicose leva a uma estimulação muito maior da resposta insulínica do que a obtida quando a glicose é infundida por via intravenosa para proporcionar níveis idênticos de glicose no sangue. Esta diferença entre as duas formas de administração de glicose tem sido atribuída às hormonas incretinas, o peptídeo semelhante ao glucagon-1 (GLP-1) e o polipéptido insulinotrópico dependente de glicose (GIP), envolvidos na eliminação da glicose absorvida, através da estimulação da secreção de insulina. De facto, os agonistas do GLP-1R e os inibidores da DPP-4 têm sido amplamente utilizados com sucesso como agentes terapêuticos no tratamento da diabetes tipo 2.
