Les incrétines sont des hormones libérées par l’intestin en réponse à l’ingestion de nutriments qui potentialisent la sécrétion d’insuline stimulée par le glucose. La recherche sur les incrétines a été motivée par l’observation que l’administration d’une charge de glucose par voie orale entraîne une stimulation beaucoup plus importante de la réponse insulinique que celle obtenue lorsque le glucose est perfusé par voie intraveineuse pour obtenir des taux de glucose sanguin identiques . Cette différence entre les deux modes d’administration du glucose a été attribuée aux hormones incrétines, le peptide-1 similaire au glucagon (GLP-1) et le polypeptide insulinotrope dépendant du glucose (GIP), qui interviennent dans l’élimination du glucose absorbé, par la stimulation de la sécrétion d’insuline. En effet, les agonistes du GLP-1R et les inhibiteurs de la DPP-4 sont des agents thérapeutiques largement utilisés et efficaces dans la prise en charge du diabète de type 2.
