Arsentrioxid (ATO) ist ein erfolgreiches Therapeutikum zur Behandlung der akuten promyelozytären Leukämie (APL), doch andere Krebsarten reagieren bei klinisch erreichbaren Dosierungen nur mäßig empfindlich auf ATO. Zudem können APL-Zellen eine Resistenz gegen ATO entwickeln. Darinaparsin (S-Dimethylarsino-GSH; Dar) ist in verschiedenen Krebsmodellen ein wirksameres Apoptose-induzierendes Arsenpräparat als ATO. Zudem liegt seine maximal tolerierte Dosis um das 35-Fache über der von ATO. Dar reichert sich in der Zelle stärker an als ATO, was zu der Hypothese führte, dass der Import von Dar zu seiner erhöhten Wirksamkeit beitragen könnte. Wir fanden heraus, dass Dar extrazellulär umgewandelt und über Cystin-/Cystein-Importsysteme in die Zelle aufgenommen wird. Wir stellten außerdem die Hypothese auf, dass die erhöhte Zytotoxizität von Dar auf eine verminderte zellschützende Reaktion zurückzuführen sein könnte. Wir konnten feststellen, dass eine ausbleibende HO-1-Aktivierung teilweise für die verstärkten Antitumoreigenschaften von Dar verantwortlich ist. Zellen reagieren auf Arsenverbindungen mit einer erhöhten Cystinaufnahme und HO-1-Expression, was zu einem Schutz vor ATO, aber zu einer Überempfindlichkeit gegenüber Dar führt. Diese Arbeit umfasst viele Aspekte der modernen molekularen Pharmakologie und der experimentellen Onkologie und soll als Inspiration für zukünftige Bemühungen zur Entwicklung von Krebsmedikamenten auf Arsenbasis dienen.
