Le cellule killer naturali (NK) fanno parte del sistema immunitario innato. Esse formano una sinapsi immunologica (IS) con cellule bersaglio infette o tumorali e le uccidono tramite recettori di morte (ligando Fas CD95 e recettore Fas CD178) o tramite il rilascio di perforina e granzima in granuli litici. Il legame Fas-FasL induce l’apoptosi. Il rilascio di perforina può provocare la lisi diretta delle cellule bersaglio (necrosi) o portare alla penetrazione dei granzimi nella cellula bersaglio, inducendo l’apoptosi. Entrambe le morti delle cellule bersaglio dipendenti dalle NK sono state descritte in letteratura, ma le analisi a livello di popolazione sono state realizzate solo attraverso test finali. La regolazione dinamica dell’apoptosi e della necrosi delle cellule bersaglio dipendenti dalle NK a livello di singola cellula non è stata ancora studiata. Per studiare i processi dinamici delle singole cellule, le cellule bersaglio che possono essere uccise dalle cellule NK umane sono state trasfettate in modo stabile con un sensore di morte cellulare. Questo test consente la classificazione dell’apoptosi e della necrosi delle singole cellule attraverso diversi modelli di fluorescenza del sensore basato su FRET.
