La tuberculose (TB) est la principale cause de décès dans le monde causée par Mycobacterium tuberculosis (M. tb). Selon l’OMS, la TB tue 5 000 personnes par jour et environ trois millions de personnes par an. Le taux de prévalence de la TB augmente de jour en jour au Pakistan. L’émergence de souches MDR de M. tb est une grande préoccupation épidémiologique. La résistance de M. tb aux antibiotiques est due à des mutations dans certains gènes. Dans cette étude, les mutations associées à la résistance à l’éthambutol (EMB) ont été déterminées par analyse RFLP. Les altérations des positions des codons 306 et 497 du gène embB ont été examinées. IS6110 est un élément d’insertion qui se trouve exclusivement chez les membres du complexe M. tuberculosis (MTBC) et qui constitue un outil de diagnostic important pour l’identification de ces espèces. L’ADN a été extrait de la culture de M. tb et soumis à la digestion par l’enzyme de restriction PvuII. Ensuite, le profil RFLP a été étudié par Southern blot pour vérifier la diversité. Le nombre de copies d’IS6110 varie dans ces isolats. Les isolats de M. tb provenant de régions proches présentent plus de similitudes que les isolats de régions éloignées.
