Tuberkulose (TB) ist die weltweit häufigste Todesursache, die durch Mycobacterium tuberculosis (M. tb) verursacht wird. Laut WHO sterben täglich 5000 Menschen und jährlich etwa drei Millionen Menschen an TB. Die Prävalenzrate von TB steigt in Pakistan von Tag zu Tag. Das Auftreten von MDR-Stämmen von M. tb ist epidemiologisch von großer Bedeutung. Die Resistenz von M. tb gegenüber Antibiotika ist auf Mutationen in bestimmten Genen zurückzuführen. In dieser Studie wurden Mutationen im Zusammenhang mit der Ethambutol (EMB)-Resistenz durch RFLP-Analyse bestimmt. Es wurden Veränderungen in den Codonpositionen 306 und 497 des embB-Gens untersucht. IS6110 ist ein Insertionselement, das ausschließlich bei Mitgliedern des M. tuberculosis-Komplexes (MTBC) vorkommt und als wichtiges diagnostisches Instrument bei der Identifizierung dieser Arten dient. Die DNA wurde aus einer M.-tb.-Kultur extrahiert und einer Verdauung durch das Restriktionsenzym PvuII unterzogen. Anschließend wurden die RFLP-Muster mithilfe von Southern Blotting untersucht, um die Diversität zu überprüfen. Die Kopienzahl von IS6110 variiert in diesen Isolaten. Isolate von M. tb aus eng verwandten Gebieten weisen mehr Ähnlichkeiten auf als Isolate aus weit entfernten Gebieten.
