This study focuses on the reactivity of 2 -aminobenzylalcohol with respect to geminalized epoxides (dicyano and cyano-ester) behaving as bi-electrophiles. The method enables highly efficient synthesis of 1,4-benzoxazepin-3-ones in very high yield, thanks to simple epoxide opening followed by cyclization. All products are characterized in minute detail by NMR, mass spectrometry and Fourier Transform Infrared spectrometry (FT-IR). With cyano-esters (2), the method enables the production of novel 1,4-benzoxazepines via an efficient one-pot reaction, without catalyst. Simple to perform, selective and achievable under mild conditions, this method opens up new prospects in medicinal chemistry, with the aim of designing new bioactive compounds.
