La tyrosine kinase ALK est exprimée, sous forme de protéine de fusion NMP/ALK, dans environ 60 % des cas de lymphome anaplasique à grandes cellules (LAGC) et dans d’autres cancers tels que le cancer du poumon non à petites cellules (CPNPC) et le neuroblastome. L’activation constitutive du domaine kinase ALK est responsable de la transformation maligne. La caractérisation structurelle du domaine kinase ALK et les informations sur les interactions médicament-protéine pourraient être très utiles dans la conception rationnelle de nouveaux médicaments spécifiques pour le traitement de l’ALCL ALK+ et d’autres tumeurs ALK+. Ce travail s’est concentré sur la production et la purification de différentes formes du domaine kinase ALK afin d’obtenir de bons candidats pour des études structurales ALK par cristallographie aux rayons X et STD-NMR. La meilleure forme de protéine recombinante a été utilisée pour cribler les inhibiteurs ALK potentiels et pour démontrer l’interaction entre le domaine kinase ALK et un nouvel inhibiteur de kinase. Les résultats de ces travaux pourraient ouvrir la voie au développement de nouveaux médicaments spécifiques pour le traitement des maladies ALK+. Ces travaux pourraient fournir un modèle méthodologique pour la production et la purification de protéines recombinantes et pour les prochaines études sur les interactions entre médicaments et protéines.
