A tirosina quinase ALK é expressa, como proteína de fusão NMP/ALK, em aproximadamente 60% dos casos de linfoma anaplásico de grandes células (ALCL) e noutros tipos de cancro, como o cancro do pulmão de células não pequenas (NSCLC) e o neuroblastoma. A ativação constitutiva do domínio quinase ALK é responsável pela transformação maligna. A caracterização estrutural do domínio da quinase ALK e as informações sobre as interações entre fármacos e proteínas podem ser muito úteis no desenho racional de novos fármacos específicos para o tratamento do ALCL ALK+ e outros tumores ALK+. Este trabalho centrou-se na produção e purificação de diferentes formas do domínio da quinase ALK para obter bons candidatos para estudos estruturais da ALK por cristalografia de raios X e STD-NMR. A melhor forma de proteína recombinante foi utilizada para selecionar potenciais inibidores da ALK e para demonstrar a interação entre o domínio da quinase ALK e um novo inibidor da quinase. Os resultados deste trabalho podem abrir caminho para o desenvolvimento de novos medicamentos específicos para o tratamento de doenças ALK+. Este trabalho pode fornecer um modelo metodológico para a produção e purificação de proteínas recombinantes e para os próximos estudos de interação medicamento-proteína.
