Kinaza tyrozynowa ALK jest wyrażana jako białko fuzyjne NMP/ALK w około 60% przypadków anaplastycznego chłoniaka wielkokomórkowego (ALCL) oraz w innych nowotworach, takich jak niedrobnokomórkowy rak płuca (NSCLC) i nerwiak niedojrzały. Za transformację złośliwą odpowiada konstytutywna aktywacja domeny kinazy ALK. Charakterystyka strukturalna domeny kinazy ALK oraz informacje na temat interakcji między lekami a białkami mogą być bardzo przydatne w racjonalnym projektowaniu nowych leków specyficznych do leczenia ALK+ ALCL i innych nowotworów ALK+. Praca ta koncentrowała się na produkcji i oczyszczaniu różnych form domeny kinazy ALK w celu uzyskania dobrych kandydatów do badań strukturalnych ALK za pomocą krystalografii rentgenowskiej i STD-NMR. Najlepsza forma białka rekombinowanego została wykorzystana do przesiewania potencjalnych inhibitorów ALK i wykazania interakcji między domeną kinazy ALK a nowym inhibitorem kinazy. Wyniki tej pracy mogą utorować drogę do opracowania nowych leków specyficznych do leczenia chorób ALK+. Praca ta może stanowić model metodologiczny dla produkcji i oczyszczania białek rekombinowanych oraz dla kolejnych badań interakcji między lekami a białkami.
