L’acquacoltura è stata introdotta nell’Africa subsahariana come innovazione per migliorare l’economia e l’alimentazione delle comunità locali. Dopo anni di investimenti locali e internazionali, l’acquacoltura continua a lottare con problemi fondamentali quali la carenza di personale, la scarsa formazione nelle tecnologie e nelle pratiche di acquacoltura, la mancanza di specie acquicole adeguate e le malattie. Questo libro si propone di descrivere la metodologia di isolamento e caratterizzazione delle proteine immunitarie (lectine e immunoglobulina mu (IgM)) dal pesce gatto africano, Clarias gariepinus e Pangasius hypophthalmus, che costituisce la base per lo sviluppo di strumenti per il monitoraggio della salute dei pesci. Sono necessarie più fasi di isolamento mediante cromatografia di affinità per isolare la proteina amiloide sierica, la proteina C-reattiva, le proteine leganti il mannosio e il galattosio. Le frazioni purificate migrano come più di una banda in Tris-tricina SDS-PAGE in presenza di beta-mercaptoetanolo. D’altra parte, le IgM isolate da P. hypophthalmus sembrano utilizzare un unico sito di riconoscimento per legarsi al fucosio e al GlcNAc. L’implicazione di questi risultati non può essere ignorata quando si considera l’utilizzo di queste lectine leganti gli zuccheri e delle IgM come strumenti per il monitoraggio della salute dei pesci.
