Il cancro è una malattia genomica complessa caratterizzata dall’accumulo di mutazioni somatiche nel corso della vita di un paziente. L’identificazione delle mutazioni somatiche driver che contribuiscono alla tumorigenesi è uno degli obiettivi principali della genomica del cancro. Grazie ai recenti progressi nelle tecnologie di sequenziamento, è diventato possibile studiare le mutazioni somatiche su scala genomica in diversi tipi di tumori. Sebbene la maggior parte degli studi di genomica del cancro si fosse precedentemente concentrata sull’identificazione delle mutazioni driver che colpiscono gli esoni, sono stati identificati diversi esempi di eventi driver all’interno delle regioni non codificanti del genoma, tra cui le mutazioni ricorrenti del promotore TERT. Tali scoperte hanno stimolato la ricerca di esempi simili di mutazioni non codificanti ricorrenti utilizzando la genomica computazionale del cancro. In questo libro presento diversi approcci computazionali volti a identificare mutazioni driver somatiche rilevanti nel cancro, con particolare attenzione alle regioni intergeniche del genoma.
