Terapia hormonalna jest pierwszą terapią celowaną, zwaną również terapią supresyjną estrogenu, często stosowaną jako terapia adiuwantowa w celu zmniejszenia ryzyka nawrotu raka po operacji lub raka, który się rozprzestrzenił. Inhibitory aromatazy, takie jak (anastrozol), są powszechnym rodzajem terapii hormonalnej; zakłócają zdolność organizmu do wytwarzania estrogenu z androgenu poprzez hamowanie enzymu aromatazy u kobiet po menopauzie z dodatnim receptorem estrogenowym raka piersi. Aromataza (cytochrom P450), zwana również syntetyzatorem estrogenów, jest enzymem odpowiedzialnym za kluczowy etap biosyntezy estrogenów. Inhibitory aromatazy odgrywają kluczową rolę w terapii hormonalnej raka piersi z dodatnim receptorem estrogenowym (ER) u kobiet po menopauzie, zakłócając zdolność organizmu do wytwarzania estrogenu z androgenów poprzez hamowanie aktywności enzymu aromatazy. Inhibitory aromatazy wykorzystują dwa różne mechanizmy blokowania działania aromatazy, a tym samym zmniejszają produkcję estrogenu.
