L’hormonothérapie est la première thérapie ciblée, également appelée thérapie de suppression des œstrogènes, souvent utilisée comme thérapie adjuvante pour réduire le risque de réapparition d’un cancer après une intervention chirurgicale ou d’un cancer qui s’est propagé. Les inhibiteurs de l’aromatase, comme l’anastrozole, sont un type courant d’hormonothérapie ; ils empêchent l’organisme de produire des œstrogènes à partir d’androgènes en supprimant l’enzyme aromatase chez les femmes ménopausées atteintes d’un cancer du sein à récepteurs d’œstrogènes positifs. L’aromatase (cytochrome P450), également appelée synthèse des œstrogènes, est une enzyme responsable d’une étape clé de la biosynthèse des œstrogènes. Les inhibiteurs de l’aromatase jouent un rôle central dans le traitement endocrinien du cancer du sein à récepteurs d’œstrogènes positifs chez les femmes ménopausées. Ils empêchent l’organisme de produire des œstrogènes à partir d’androgènes en supprimant l’activité de l’enzyme aromatase. Les inhibiteurs de l’aromatase utilisent deux mécanismes distincts pour bloquer l’action de l’aromatase et réduire ainsi la production d’œstrogènes.
