Os iões de cálcio (Ca2+) são moléculas de sinalização essenciais e devem ser cuidadosamente regulados para preservar a especificidade da sua função. Uma proteína de sinalização de Ca2+ indispensável é o canal de libertação de cálcio do recetor de rianodina (RyR). O RyR1 é essencial para a contração muscular, e as mutações do RyR1 podem levar a doenças musculares graves, como a hipertermia maligna (HM). Recentemente, foi desenvolvido um modelo de ratinho da HM humana (Y522S-RyR1). No músculo esquelético, o Y522S-RyR1 apresenta uma sensibilidade aumentada à ativação, resultando em fuga de Ca2+, desorganização mitocondrial e stress celular. Embora a expressão de RyR1 seja limitada no cérebro, o cerebelo pode ser particularmente vulnerável a esta doença porque RyR1 é altamente expresso em neurónios Purkinje. Neste estudo, o Y522S-RyR1 nas células de Purkinje apresenta um limiar de ativação mais baixo, mas não causa stress e danos celulares graves. A investigação futura de mecanismos compensatórios para o aumento da libertação de Ca2+ nas células de Purkinje poderá ter valor terapêutico para muitas doenças neurológicas e musculares.
