Der Samen der Jackfrucht ist fest und wachsartig, oval, länglich oder länglich-ellipsenförmig. Jeder Samen ist 2-4 x 1-2 cm groß und wiegt 2,5-14 g. Er hat eine ledrige Samenschale. Viele Blüten am Blütenstand produzieren keine Samen; normalerweise befinden sich 100-500 Samen in jeder Frucht. In den meisten Ländern, in denen Jackfrüchte angebaut werden, wird die Vermehrung über Samen praktiziert. Die Keimfähigkeit von Jackfruchtsamen geht sehr schnell verloren. Selbst eine Verzögerung von ein oder zwei Wochen beim Pflanzen führt zu einer schlechten Keimung. Jackfruchtsamen gelten als schwer keimfähig und können bei Raumtemperatur nur wenige Tage oder Wochen gelagert werden.
