Negli ultimi decenni, la prevalenza delle infezioni fungine invasive (IFI) è aumentata costantemente in tutto il mondo. I pazienti immunodepressi presentano forme cliniche gravi di queste infezioni, spesso letali, e sono più suscettibili alle infezioni fungine opportunistiche. Le IFI sono state associate a un’elevata morbilità e mortalità, in parte a causa dei limiti delle terapie antifungine disponibili. La ricerca di terapie alternative e/o lo sviluppo di farmaci più specifici è una sfida che deve essere affrontata. Analisi in silico e mining manuale hanno selezionato 10 geni presenti nei funghi patogeni e assenti nel genoma umano. Ci siamo concentrati sul TRR1 del Paracoccidioides lutzii che codifica per la tioredossina reduttasi. Qui viene presentata una strategia di screening assistita da computer contro la proteina TRR1. Tra le molecole testate, 3 presentano attività inibitoria nei test antifungini contro i funghi patogeni. A conferma di questi risultati, le molecole hanno attività inibitoria contro l’enzima ricombinante purificato TRR1 nei test biochimici. Questi risultati forniscono una tecnica promettente per lo sviluppo di farmaci nuovi e innovativi.
