W ciągu ostatnich kilku dekad na całym świecie obserwuje się stały wzrost częstości występowania inwazyjnych zakażeń grzybiczych (IFI). Pacjenci z obniżoną odpornością cierpią na ciężkie postacie kliniczne tych zakażeń, które często kończą się zgonem, a ponadto są bardziej podatni na oportunistyczne zakażenia grzybicze. IFI wiążą się z wysoką zachorowalnością i śmiertelnością, częściowo z powodu ograniczeń dostępnych terapii przeciwgrzybiczych. Poszukiwanie alternatywnych terapii i/lub opracowanie bardziej specyficznych leków stanowi wyzwanie, któremu należy sprostać. Analizy in silico oraz ręczne przeszukiwanie danych pozwoliły wyłonić 10 genów obecnych w grzybach chorobotwórczych, a nieobecnych w genomie człowieka. Skupiliśmy się na TRR1 z Paracoccidioides lutzii, który koduje reduktazę tioredoksyny. W niniejszym artykule przedstawiono strategię komputerowego przesiewania przeciwko białku TRR1. Spośród badanych cząsteczek 3 wykazują aktywność hamującą w testach przeciwgrzybiczych przeciwko grzybom chorobotwórczym. Potwierdzając te wyniki, cząsteczki wykazują aktywność hamującą wobec oczyszczonego rekombinowanego enzymu TRR1 w testach biochemicznych. Wyniki te stanowią obiecującą technikę w rozwoju nowych i innowacyjnych leków.
