Le fer est le métal de transition le plus abondant dans les systèmes biologiques ; il est indispensable à de nombreuses réactions métaboliques clés, notamment la synthèse de l’ADN, le transport de l’oxygène et la respiration cellulaire. La distribution du fer dans l’organisme est rigoureusement régulée afin d’empêcher la formation d’espèces réactives de l’oxygène cytotoxiques. Les recherches décrites dans cet ouvrage visaient à comprendre les mécanismes et les maladies associés au métabolisme du fer chez les mammifères. Plus précisément, le chapitre 3 examine l’activité homéostatique systémique de l’hepcidine, l’hormone du métabolisme du fer, lorsqu’elle est liée à la protéine de transport sanguin, l’α2-macroglobuline ; le chapitre 4 examine le ou les mécanismes moléculaires impliqués dans la perturbation du métabolisme cellulaire du fer qui entraîne une accumulation de fer dans les mitochondries dans l’ataxie de Friedreich ; et le chapitre 5 explore les liens moléculaires entre la dérégulation du fer mitochondrial et les anomalies cardiaques mortelles observées dans l’ataxie de Friedreich.
