Ziel dieser Studie war die Entwicklung einer Multiplex-RT-PCR mit hochauflösender Schmelzkurvenanalyse (HRM) für den schnellen und genauen Nachweis von fünf Carbapenemase-housekeeping-Gen-16S-rRNA. Sechs Primerpaare und RT-PCR-Bedingungen wurden mit der Beacon Designer Software entworfen. Die Identifizierung der Gene erfolgte durch Schmelzkurven und Temperaturen mit Multiplex RT-PCR unter Verwendung eines speziellen HRM-Reagens. Dies ist der erste Assay, der die wichtigsten Carbapenemase-Gene in einem Zwei-Röhrchen-Assay nachweist und den gleichzeitigen Nachweis von 5 der wichtigsten Marker für die Resistenz gegen Cephalosporine und Carbapeneme der dritten Generation ermöglicht.
