Kürzlich wurde über die umstrittene Rolle von Cannabinoidrezeptoren in der Pathophysiologie der Parkinson-Krankheit berichtet. In der vorliegenden Studie haben wir die Rolle von CB1-Rezeptoren im motorischen Verhalten untersucht. Zur Erzeugung stabiler motorischer Defizite bei Ratten wurde eine wiederholte intranigrale Infusion (Tag 1, 7, 14) von MPTP durchgeführt. Der CB1-Rezeptoragonist Noladin-Ether und der Antagonist AM 251 wurden den mit MPTP behandelten Ratten allein oder in Kombination verabreicht. Zur Beurteilung des motorischen Verhaltens wurden Tests zur Griffkraft, zur spontanen Bewegungsaktivität und zum Gehen auf einem schmalen Balken durchgeführt. Oxidativer Stress und die Konzentrationen proinflammatorischer Zytokine wurden in Homogenaten des Striatums bestimmt. Die mit MPTP behandelten Ratten zeigten stabile motorische Defizite sowie einen Anstieg des oxidativen Stresses und der Zytokinwerte. AM 251 milderte die MPTP-induzierte motorische Dysfunktion, den oxidativen Stress und die Neuroinflammation bei Ratten. Umgekehrt verstärkte Noladin-Ether die MPTP-vermittelte Neurotoxizität, die durch eine Vorbehandlung mit AM 251 signifikant abgeschwächt wurde.
