Recentemente, foi relatado o papel controverso dos recetores canabinóides na fisiopatologia da DP. No presente estudo, investigámos o papel dos recetores CB1 no comportamento motor. Foram realizadas infusões intranigrais repetidas (dias 1, 7 e 14) de MPTP para induzir défices motores estáveis em ratos. O agonista do receptor CB1, éter de noladina, e o antagonista AM 251 foram administrados isoladamente ou em combinação em ratos que receberam infusão de MPTP. Foram utilizados testes de força de preensão, atividade locomotora espontânea e caminhada em viga estreita para avaliar os comportamentos motores. Os níveis de stress oxidativo e de citocinas pró-inflamatórias foram determinados em homogeneizados do estriado cerebral. Os ratos que receberam infusão de MPTP apresentaram défice motor estável e elevação dos níveis de stress oxidativo e de citocinas. O AM 251 atenuou a disfunção motora induzida pelo MPTP, o stress oxidativo e a neuroinflamação em ratos. Por outro lado, o éter de noladina sinergizou a neurotoxicidade mediada pelo MPTP, que foi significativamente atenuada pelo pré-tratamento com AM 251. Os resultados acima sugerem que a ativação sustentada dos recetores CB1 pode desempenhar um papel crucial no desenvolvimento da disfunção motora e, portanto, o bloqueio dos recetores CB1 seria terapêutico na DP.
