La présente étude a été menée sur le phasol à grappes afin d’étudier l’effet d’une salinité à dominante chlorure sur la croissance et d’explorer la possibilité d’induire une tolérance à la salinité par l’utilisation de l’EMS. L’accumulation de sucres solubles et de proline libre dans les feuilles en présence de salinité indique, dans une certaine mesure, un ajustement osmotique des feuilles, mais la diminution de la teneur en protéines et en amidon est probablement due à la toxicité ionique. Le nœud cotylédonaire s’est avéré être le meilleur explant et le milieu MSB5 + 2 mg/l de 2,4-D et 1 mg/l de BAP a favorisé la meilleure croissance du cal. Une concentration de 200 mM de NaCl s’est avérée létale. La culture de callos traités à l’EMS sur un milieu enrichi en NaCl n’a montré aucune croissance significative du callus jusqu’à 2,0 h de traitement à l’EMS. Une augmentation de la durée du traitement à l’EMS au-delà de 2,0 h a favorisé la croissance du callus, qui a augmenté jusqu’à 3,0 h. En général, la teneur en amidon, en protéines, en proline et en sodium a augmenté sur le milieu enrichi en sel par rapport au contrôle respectif, tandis que la teneur en chlorure a diminué par rapport au contrôle respectif. Le NaCl a induit de nouvelles bandes de 57,54, 28,18, 26,92 et 20,89 kDa. La culture de ces callosités sur un milieu enrichi en sel a révélé la présence de bandes de polypeptides de 67,61, 53,70, 50,12, 28,84, 20,89 et 18,62 kDa.
