La germinazione dei semi è controllata da segnali ambientali, tra cui la luce e i fitormoni endogeni. L’acido abscissico (ABA) inibisce, mentre la gibberellina (GA) promuove rispettivamente la germinazione e lo sviluppo iniziale delle piantine. Qui riportiamo che ZFP3, una proteina nucleare C2H2 con dito di zinco, agisce come regolatore negativo della germinazione soppressa dall’ABA. Di conseguenza, la sovraespressione regolata di ZFP3 e dei fattori con dito di zinco strettamente correlati ZFP1, ZFP4, ZFP6 e ZFP7 conferisce insensibilità all’ABA alla germinazione dei semi, mentre il doppio mutante zfp3 zfp4 mostra una maggiore sensibilità all’ABA. La ridotta espressione di diversi geni indotti dall’ABA, come RAB18 e il fattore di trascrizione ABI4 nelle piantine ZFP3ox, suggerisce che ZFP3 regola negativamente la segnalazione dell’ABA. L’analisi delle piante ZFP3ox ha rivelato molteplici alterazioni fenotipiche, quali portamento seminarca, difetti di fertilità e maggiore sensibilità dell’allungamento dell’ipocotile alla luce rossa, ma non alla luce rossa lontana o blu. L’analisi delle interazioni genetiche con i mutanti fitocromo e abi indica che ZFP3 regola le risposte alla luce e all’ABA durante la germinazione e lo sviluppo iniziale delle piantine.
