Die Studie konzentriert sich auf die Entwicklung und Bewertung von bukkalen mukoadhäsiven Tizanidin-Pflastern als neuartiger Ansatz zur Verbesserung der systemischen Verabreichung von Medikamenten. Tizanidin weist aufgrund eines umfangreichen First-Pass-Metabolismus eine geringe orale Bioverfügbarkeit auf, so dass der bukkale Weg eine geeignete Alternative für eine verbesserte Absorption darstellt. Die Pflaster wurden unter Verwendung von Polymeren wie Polyvinylalkohol (PVA) und Polyvinylpyrrolidon (PVP) durch das Lösungsmittelgießverfahren hergestellt und auf verschiedene physikochemische und mechanische Eigenschaften hin untersucht, darunter Dicke, Zugfestigkeit, Quellungsindex und Gleichmäßigkeit des Wirkstoffgehalts. In-vitro-Freisetzungsstudien zeigten eine kontrollierte Wirkstofffreisetzung zwischen 71 % und 97 % innerhalb von 90 Minuten, die einer diffusionskontrollierten Kinetik folgt. In-vivo-Studien an Kaninchen und menschlichen Probanden bestätigten eine verbesserte Arzneimittelabsorption und eine gute Korrelation mit den In-vitro-Ergebnissen. Insgesamt deuten die Ergebnisse darauf hin, dass bukkale mukoadhäsive Pflaster eine vielversprechende und wirksame Alternative zur oralen Verabreichung darstellen, die eine erhöhte Bioverfügbarkeit, eine kontrollierte Freisetzung und verbesserte therapeutische Ergebnisse bietet.
