Le cancer est la deuxième cause de décès. Son étiologie reflète très probablement la contribution de nombreux facteurs. Notre étude démontre que la progression maligne du cancer du sein est associée à une dépendance accrue à l’antiporteur xc-cystine/glutamate. Nous fournissons des preuves suggérant qu’un médicament anti-inflammatoire très couramment utilisé, la sulfasalazine, antagonise sélectivement l’antiporteur xc- et inhibe la prolifération des cellules épithéliales mammaires. Les études sur la relation structure-activité ont identifié diverses fractions de la sulfasalazine qui pourraient être impliquées dans l’antagonisme de l’antiporteur xc-. La suppression de l’absorption de la cystine par la sulfasalazine a entraîné une déplétion du glutathion intracellulaire (GSH) de manière dépendante du temps et de la concentration. De plus, les données suggèrent que la sulfasalazine pourrait être un inhibiteur de la pompe d’efflux des médicaments, la protéine de résistance associée à la multirésistance. En conclusion, nous avons identifié un nouveau marqueur de la progression tumorale, l’antiporteur xc, et révélé le rôle potentiel du SASP dans le traitement du cancer.
