Metforminhydrochlorid ist das am häufigsten verwendete orale Antidiabetikum. Bei Diabetikern muss die Metformin-Konzentration im Blut regelmäßig gemessen werden, um eine Metformin-Toxizität zu verhindern und die Einhaltung der Medikamenteneinnahme zu überprüfen. Zur Gewinnung von Proben für die Analyse von Metformin wurde die Technik der getrockneten Blutstropfen (DBS) verwendet. Es wurde ein einfaches HPLC-Verfahren zur quantitativen Bestimmung von Metformin im menschlichen Plasma entwickelt. Das Verfahren umfasst eine C-18-Stationärphase unter Verwendung einer 34%igen Acetonitril- und einer 66%igen wässrigen Phase. Die wässrige Phase enthielt 10 mM Kaliumphosphat (KH2PO4) und 10 mM Natriumdodecylsulfat (SDS), der pH-Wert betrug 5,3, ein UV-Detektor (eingestellt auf 234 nm) wurde verwendet. Das Metformin wurde erfolgreich aus getrockneten Blutflecken durch eine kostengünstige, einfache, einstufige Flüssigextraktion mit 100 % Methanol für 2 Minuten extrahiert. Es wurden nur 10 μl Blut benötigt. Die durchschnittliche Wiederfindungsrate von Metformin aus DBS betrug 95 %.
