O ARN mensageiro tornou-se o foco da investigação em medicina molecular, uma vez que é uma molécula não tóxica que permite a expressão transitória de caproteínas em praticamente todos os tipos de células, incluindo as células dendríticas. Os princípios subjacentes às vacinas de ácidos nucleicos estão enraizados no dogma central de Watson e Crick - que o ADN é transcrito em ARNm, que por sua vez é traduzido em proteínas. O ARNm é produzido sinteticamente e formulado em vacinas. Na síntese de vacinas de ARNm, as transcrições de ARNm são purificadas por cromatografia líquida de alta eficiência (HPLC) para remover contaminantes e reagentes. Os ARNm são traduzidos em antigénios correspondentes depois de serem inoculados nas células hospedeiras, imitando a imunidade humoral e a imunidade celular semelhantes a uma infeção por vírus. Com a ajuda da vacina de ARNm, é possível aumentar a reatividade dos antigénios. As vacinas de ARNm optimizadas devem cumprir o requisito de que a imunidade inata seja totalmente activada para iniciar a imunidade adaptativa. A primeira vacina candidata lançada em ensaios clínicos é uma vacina de ARNm administrada através de nanopartículas lipídicas. Esta vantagem determina a rápida resposta imunitária após a administração do ARNm. Vacina de ARNm amplamente utilizada em doenças infecciosas.
