Nella ricerca e nello sviluppo di sistemi di somministrazione orale di farmaci a velocità controllata, negli ultimi anni sono stati fatti progressi scientifici e tecnologici superando le avversità fisiologiche, come i brevi tempi di residenza gastrica (GRT) e i tempi di svuotamento gastrico irregolari (GET). Diversi metodi, compresi i sistemi di rilascio di farmaci galleggianti (FDDS), noti anche come sistemi idrodinamicamente bilanciati (HBS), sistemi di rigonfiamento ed espansione, sistemi polimerici bioadesivi, sistemi di forma modificata, sistemi ad alta densità e altri dispositivi a svuotamento gastrico ritardato, sono attualmente utilizzati nel prolungamento del GRT. Le varie strategie utilizzate nello sviluppo di FDDS costruendo il meccanismo di galleggiamento basato sulla forma effervescente e non effervescente delle compresse galleggianti. FDDS è un metodo di consegna di farmaci localmente attivi con una piccola finestra di assorbimento nel tratto gastrointestinale superiore, instabile nell’ambiente intestinale inferiore, e con valori di pH più alti che hanno una bassa solubilità. I recenti progressi nei sistemi di consegna dei farmaci galleggianti includono variabili fisiologiche e di formulazione che influenzano il tempo di ritenzione gastrica, approcci alla formulazione di sistemi galleggianti per unità singole e unità multiple.
