L’adenocarcinoma del colon è il tipo più comune di cancro gastrointestinale. Per questo motivo, è stato eseguito il sequenziamento del DNA di frammenti di cDNA su una linea cellulare Caco-2. Questo esperimento è stato eseguito per trovare potenziali marcatori biologici della cellula cancerosa o dell’enterocita normale, che potrebbero essere utili nella diagnosi della malattia. Il sequenziamento del DNA è stato eseguito con il metodo della terminazione di catena con ddNTP marcati e la loro lettura con un sequenziatore di DNA (Beckman Coulter CEQ 2000). Delle 11 sequenze ottenute con successo, 5 frammenti acquisiti sono stati parzialmente abbinati a sequenze di DNA umano e hanno rivelato che i geni completi, a cui appartengono questi frammenti, codificano 3 diverse proteine umane. Queste proteine sono la Guanine Nucleotide-Binding Protein Subunit Beta-2-Like 1, la Chromodomain Helicase DNA Binding Protein 4 e la Centromere protein J. È stato riscontrato che esse contribuiscono o inibiscono i fenotipi cancerosi delle cellule, il che potrebbe essere spiegato dalla quantità di proteine presenti nella cellula e dal livello di mutazioni che possiedono. Questa ricerca rivela la necessità di ulteriori ricerche sulle linee cellulari tumorali e sul loro sequenziamento del DNA.
