Ein Säugetierkörper besteht aus mehr als 200 Zelltypen, die fast alle das gleiche Genom, aber unterschiedliche Morphologie und Funktionen haben. Diese Unterschiede sind auf die zellspezifische Genregulationsmaschinerie zurückzuführen. Das Genom von Säugetieren weist eine komplexe Organisation von regulatorischen Sequenzen auf, die mit verschiedenen regulatorischen Elementen verbunden sind. Die lineare DNA im Zellkern ist auf sehr komplexe Weise gefaltet, und diese Faltung bringt weit voneinander entfernte Regionen näher zueinander. Die Kombination verschiedener DNA-bindender Proteine wie Transkriptionsfaktoren und Histone, epigenetische Markierungen wie Genposition und dynamische Modifikation von DNA und Histonen sowie die dreidimensionale Organisation der regulatorischen Landschaft interagieren auf durchdringende und zellspezifische Weise, um komplexe raum-zeitliche Expressionsmuster zu erzeugen. Chromatin-Immunpräzipitation (ChIP seq) und Chromosome Conformation Capture (Hi C und 5C) können mit der Shotgun-Sequenzierung des gesamten Transkriptoms (RNA-Seq) kombiniert werden, um das räumliche Muster der Bindung von regulatorischen Elementen an ihre Zielgene zu entschlüsseln.
