Gli scaffold 3D di chitosano (CHI), fibroina (SF) e idrossiapatite (HA) sono biocompatibili e interessanti per applicazioni nell’ingegneria tissutale ossea. Pertanto, l’obiettivo era quello di preparare, caratterizzare e valutare il comportamento in vitro degli scaffold 3D di CHI, CHI/SF, CHI/SF/HA. I biomateriali sono stati prodotti a partire da una soluzione di CHI, CHI/SF e CHI/SF/HA in acido acetico e, dopo la realizzazione dei processi di produzione, sono stati sottoposti a test in vitro con cellule SaOS-2 per 7, 14 e 21 giorni. Gli scaffold sono stati caratterizzati mediante diffrazione dei raggi X, spettroscopia infrarossa con trasformata di Fourier con riflettanza totale attenuata, analisi termogravimetrica, calorimetria differenziale esplorativa, microscopia elettronica a scansione, spettroscopia a energia dispersiva e sottoposti a test per verificare la porosità, la dimensione dei pori e l’angolo di contatto. La fattibilità e l’attività cellulare sono state valutate mediante riduzione dell’MTT e rilevamento dell’attività della fosfatasi alcalina. Gli scaffold 3D sono stati considerati atossici e in grado di promuovere l’adesione, la proliferazione e la differenziazione cellulare, potendo essere utilizzati nella terapia di riparazione dei difetti ossei.
