A aterosclerose é a patologia subjacente a doenças como a doença arterial coronária, resultando em mortalidade e morbidade geral elevadas. É uma doença inflamatória crónica da parede arterial, na qual o sistema imunitário inato tem uma contribuição fundamental. O receptor Toll-like 9 (TLR9) é um importante receptor imunitário intracelular inato que tem atraído grande interesse e está atualmente a ser testado clinicamente como alvo terapêutico de doenças inflamatórias. Utilizando ratos com deficiência de apolipoproteína E alimentados com uma dieta rica em gordura durante períodos de 8 a 20 semanas como modelo de aterosclerose, utilizámos uma abordagem genética (deleção de TLR9) e farmacológica (agonista de TLR9) para demonstrar um papel protetor do TLR9 na aterosclerose. Descrevemos a modulação do número de células e do fenótipo inflamatório das células T CD4+ como um mecanismo potencial. Além disso, identificámos uma abordagem terapêutica nova e atraente para atenuar a progressão da doença aterosclerótica com base na utilização de oligodesoxinucleótidos CpG ativadores do TLR9.
