O Papilomavírus Humano (HPV) é um vírus de ADN que infeta as células epiteliais da pele e das membranas mucosas. Os tipos de HPV de alto risco, nomeadamente o HPV-16 e o HPV-18, são reconhecidos como importantes fatores de risco no desenvolvimento de cancros orais e orofaríngeos. Nos últimos anos, a incidência de cancros orais associados ao HPV tem aumentado a nível global, o que realça a necessidade de uma maior sensibilização e investigação nesta área. O potencial carcinogénico do HPV deve-se principalmente às oncoproteínas virais E6 e E7. Estas proteínas perturbam a regulação normal do ciclo celular, inativando as proteínas supressoras de tumores p53 e retinoblastoma (Rb). Como resultado, as células infetadas sofrem uma proliferação descontrolada, acumulam mutações genéticas e podem, eventualmente, transformar-se em células malignas. A infeção persistente por estirpes de HPV de alto risco é, por conseguinte, um fator significativo na carcinogénese oral. Os cancros orais HPV-positivos diferem dos cancros orais tradicionais associados ao consumo de tabaco e álcool. Ocorrem frequentemente em indivíduos mais jovens e caracterizam-se por características moleculares e clínicas distintas. É importante referir que os doentes com cancros orais relacionados com o HPV apresentam, geralmente, uma melhor resposta ao tratamento
