Stosowanie NLPZ drogą ogólnoustrojową może wywoływać działania niepożądane, ponieważ wymagane są duże dawki. W przypadku podawania doustnego pacjent może odczuwać działania niepożądane, takie jak owrzodzenia żołądkowo-jelitowe i nietolerancja, ze względu na kwaśny charakter NLPZ. Ponadto lek podawany drogą ogólnoustrojową może być metabolizowany w wątrobie. W związku z tym dochodzi do utraty leku. Aceclofenac jest niesteroidowym lekiem przeciwzapalnym, który jest pochodną kwasu fenylooctowego o skutecznych właściwościach przeciwzapalnych i przeciwbólowych. Jest to selektywny inhibitor COX-2, stosowany głównie w łagodzeniu bólu i stanów zapalnych w chorobie zwyrodnieniowej stawów, reumatoidalnym zapaleniu stawów i zesztywniającym zapaleniu stawów kręgosłupa. W przypadku aceklofenaku wybrano podawanie przezskórne, aby uniknąć metabolizmu pierwszego przejścia w wątrobie, przezwyciężyć podrażnienie przewodu pokarmowego, ponieważ lek ma kwaśny charakter, oraz zlokalizować lek w miejscu choroby poprzez podawanie przezskórne.
