Die Polymerase-Kettenreaktion (PCR) dient in erster Linie der Vervielfältigung eines DNA-Abschnitts, wobei Millionen von Kopien des ausgewählten Abschnitts entstehen. Das Klonen von Genen zielt darauf ab, ein ausgewähltes Gen in Bakterien einzuführen, die untereinander identisch sind, und diese Technik erfordert das Vorhandensein eines Klonierungsvektors, der in der Lage ist, ein in ihn geklontes DNA-Fragment zu amplifizieren. Das E-Gen des Dengue-Virus (DENV) wird aufgrund der Bedeutung des E-Proteins am häufigsten für molekulare Studien verwendet. Ziel dieser Studie ist es, Techniken zur Amplifikation des E-Gens von DENV-2 und DENV-4 zu entwickeln. Das DNA-Molekül wurde vom FIOCRUZ in Recife bezogen; die Primer wurden so ausgewählt, dass sie das E-Gen enthalten. Die Fragmente wurden mittels Agarosegel-Elektrophorese aufgetrennt. Die PCR-Amplifikation des E-Gens verlief zufriedenstellend. Diese Techniken sind für die Gewinnung rekombinanter DNA bei der Klonierung und Expression des Dengue-E-Proteins unerlässlich.
