Die Aufrechterhaltung eines funktionsfähigen, lebenslangen Gewebes erfordert einen kontinuierlichen Zellumsatz. Die Fluktuationsrate wird daher sorgfältig mit funktionsgestörten und absterbenden Zellen ausbalanciert. Der physiologische Zellumsatz hängt hauptsächlich von den ansässigen, terminal differenzierten Zellen oder Stammzellen ab. Endgültig differenzierte Zellen können eine Replikationstendenz haben, die es ihnen ermöglicht, proliferierende Vorläuferzellen zu vermehren. Die meisten Organe und Gewebe haben ihre eigenen Stamm- oder Vorläuferzellen, die die Fähigkeit zur Differenzierung behalten. Die Ausrichtung der Stammzellen auf ihren Bestimmungsort hängt von ihrer Nische und ihrer Mikroumgebung ab. Bestimmte Faktoren üben ihre Wirkung auf Stammzellen aus, um die spezifische Genexpression zu steuern, die wiederum die Zellvermehrung und -differenzierung fördert (Sanchez Alvarado und Yamanaka, 2014).
