Libro primero de las epístolas familiares es un conjunto de textos publicados en dos libros que contienen ochenta y cinco cartas, veintidós razonamientos, discursos y sermones. El primero libro apareció en 1539, y el segundo en 1542. Las Epístolas tratan sobre temas variados: consejos a viudas, y hasta una censura a una sobrina desesperada por la muerte de su perra. Contiene sátiras, chistes, anécdotas, transcripciones y comentarios diversos. Hay epístolas de interés político y también histórico, otras hablan de la influencia de los humores en las enfermedades, de los enojos que hacen padecer a los enamorados, del tocado de las damas, y en otras se comentan textos sagrados. Cada epístola esta dirigida a personas de su tiempo, entre otros: a Alonso Manrique, arzobispo de Sevilla; don Jerónimo Vique, embajador; don Gonzalo Fernández de Córdoba, Gran Capitán; y a mosé Puché. Ellas muestran un panorama de la vida social, política, jurídica y religiosa, del reinado de Carlos V.
