Le maintien d’un tissu fonctionnel tout au long de la vie nécessite un renouvellement cellulaire continu. Le taux de renouvellement est donc soigneusement équilibré avec les cellules dysfonctionnelles et mourantes. Le renouvellement cellulaire physiologique dépend principalement des cellules terminalement différenciées ou des cellules souches résidentes. Les cellules terminalement différenciées peuvent avoir une tendance à la réplication qui leur permet de propager des précurseurs en prolifération. La plupart des organes et des tissus possèdent leurs propres cellules souches ou progénitrices qui conservent une capacité de différenciation. L’orientation des cellules souches vers la destination cible dépend de leur niche et de leur microenvironnement. Certains facteurs exercent leur effet sur les cellules souches pour guider l’expression de gènes spécifiques qui, à leur tour, favorisent la propagation et la différenciation des cellules (Sanchez Alvarado et Yamanaka, 2014).
