L’ADN est une cible privilégiée des médicaments dans la cellule. Le mode d’action de nombreux agents chimiothérapeutiques serait dû à leur capacité à s’intercaler entre les paires de bases de l’ADN à double hélice. On pense généralement que les atraquinones exercent leurs actions biologiques en se liant à l’ADN, puis en interférant avec la réplication de l’ADN, la transcription et l’inhibition de l’expression des gènes. La conception de petites molécules médicamenteuses qui ciblent sélectivement l’ADN, avec des affinités de liaison élevées, a conduit à la découverte de nombreux médicaments anticancéreux, antibiotiques et antiviraux. La plupart des molécules ciblant l’ADN commencent par se lier à l’ADN à double hélice de manière non covalente, puis peuvent évoluer vers une liaison covalente. La liaison non covalente peut inclure l’empilement π, la liaison hydrogène, l’électrostatique, le transfert de charge et les interactions hydrophobes. Toutes ces interactions peuvent contribuer au mécanisme d’interaction médicament/ADN, de sorte que l’objectif principal de cette étude est d’explorer l’interaction dominante. Ces informations sont cruciales pour la conception et le développement de nouveaux médicaments.
