Die Genomtechnik steht kurz vor dem Beginn ihres goldenen Zeitalters. Neue und aufregende Werkzeuge tauchen auf und erweitern das Arsenal. Nach TALENs und Zing-Fingern kommt nun ein neues und vielversprechendes Werkzeug hinzu. Die neueste Ergänzung ist das CRISPR-System. Ein System, das zuerst bei Streptococcus thermophilus entdeckt wurde und dessen natürliche Rolle die sequenzspezifische adaptive Immunität gegen fremde DNA in Bakterien ist. Im vorliegenden Bericht wird das CRISPR Cas9-System vom Typ II verwendet, um das in Lymphozyten exprimierte HPRT1-Gen auszuschalten, um ein Konzept zu testen. Als Selektionsmedium wird HAT-Medium in Kombination mit Thioguanin verwendet, was ein schnelles und einfaches Auslese-Screeningverfahren ermöglicht. Um zu überprüfen, ob ein erfolgreiches Editing-Ereignis in den Säugetierzellen stattgefunden hat, wird ein DNA-Spaltungskit in Kombination mit einem enzymatischen Assay verwendet.
