Il salbutamolo solfato (SS), un simpaticomimetico ad azione diretta utilizzato per il trattamento dell’asma acuta e cronica, è stato formulato come gel in situ a base di Gelerite®. I gel in situ a base di SS (2% p/p) sono stati preparati a concentrazioni di gomma di gellan dello 0,2%, 0,4% e 0,8% p/v con metilparaben (0,05%) come conservante. Il rilascio in vitro in fluido nasale simulato ha mostrato un rilascio relativamente rapido e la cinetica di rilascio ha seguito il modello cinetico di diffusione. Il test di mucoadesività in vivo eseguito sui ratti ha mostrato un tempo di trasporto mucociliare prolungato da 5 minuti (soluzione di controllo) a 16,5 e 19,5 minuti per le formulazioni contenenti rispettivamente lo 0,4% e lo 0,8%. I gel in situ hanno dimostrato un’adeguata sicurezza per la mucosa nasale dei ratti. Il gel in situ allo 0,4% è rimasto liquido per 6 mesi senza che si verificassero torbidità o gelificazione alle condizioni di temperatura di conservazione. L’analisi statistica ha rivelato una differenza non significativa nell’uniformità del contenuto o nel rilascio rispetto a quelli freschi. Il gel in situ è un approccio promettente per la somministrazione intranasale di SS, che combina il vantaggio di una soluzione, la comodità di somministrazione e il dosaggio esatto con un tempo di permanenza favorevole e un previsto miglioramento dell’assorbimento del farmaco.
