Au cours des dernières décennies, les infections fongiques (IFI) ont considérablement augmenté, constituant une menace importante pour la santé humaine. Candida albicans, Cryptococcus neoformans et Aspergillus fumigatus sont les principaux agents pathogènes responsables de la majorité des IFI en milieu clinique. Malgré les progrès de la thérapie antifongique, les IFI restent une cause majeure de morbidité et de mortalité, en particulier chez les patients immunodéprimés et infectés par le VIH. Les principales familles thérapeutiques pour les infections fongiques comprennent les polyènes, les fluoropyrimidines, les azoles, les allylamines et les échinocandines. Toutefois, ces médicaments antifongiques sont confrontés à des problèmes tels que la faible solubilité dans l’eau, la toxicité, l’activité à spectre étroit, la faible biodisponibilité et, surtout, l’émergence d’une résistance aux médicaments fongiques. Pour répondre au besoin médical urgent d’agents antifongiques améliorés, ce livre explore l’application des méthodes de conception de médicaments assistée par ordinateur (CADD).
