In den letzten Jahrzehnten hat die Zahl der Pilzinfektionen (IFI) erheblich zugenommen und stellt eine erhebliche Gefahr für die menschliche Gesundheit dar. Candida albicans, Cryptococcus neoformans und Aspergillus fumigatus sind die vorherrschenden Erreger, die für die Mehrzahl der IFI im klinischen Bereich verantwortlich sind. Trotz Fortschritten in der antimykotischen Therapie sind Pilzinfektionen nach wie vor eine der Hauptursachen für Morbidität und Mortalität, insbesondere bei immungeschwächten und HIV-infizierten Patienten. Zu den primären therapeutischen Familien für Pilzinfektionen gehören Polyene, Fluoropyrimidine, Azole, Allylamine und Echinocandine. Diese Antimykotika stehen jedoch vor Herausforderungen wie geringer Wasserlöslichkeit, Toxizität, engem Wirkungsspektrum, geringer Bioverfügbarkeit und vor allem dem Auftreten von Pilzresistenzen. Um den dringenden medizinischen Bedarf an verbesserten Antimykotika zu decken, untersucht dieses Buch die Anwendung von CADD-Methoden (Computer-Aided Drug Design)
