L-Glutamin-(Asparagin-)ase ist ein Amidohydrolase-Enzym, das die Hydrolyse von L-Asparagin (L-asn) und L-Glutamin (L-gln) zu L-Aspartat (L-asp) bzw. L-Glutamat (L-glu) katalysiert und dabei Ammoniak (NH3) freisetzt.Die Aktivität von L-Glutamin-(industriell). So kann es beispielsweise für die Herstellung von Spezialchemikalien wie Thiamin und Glutamin verwendet werden oder als Biosensor zur Überwachung des L-Glutaminspiegels dienen. In der Klinik gilt die Senkung des Asparagin- und/oder Glutaminspiegels durch die Verabreichung von bakterieller Asparaginase und/oder Glutaminase oder durch die Verwendung ihrer Antagonisten als gängige Therapie für Patienten mit akuter lymphatischer Leukämie. Im Gegensatz dazu fördert die Supplementierung mit Asparagin und Glutamin bei menschlichen Dickdarmkrebs-Zelllinien einen bösartigeren und weniger differenzierten Phänotyp.
