La giunzione neuromuscolare subisce un processo di maturazione fisiologica dopo il contatto iniziale tra nervo e muscolo, caratterizzato da un aumento spontaneo dell’ampiezza della corrente sinaptica e da una diminuzione del tempo di salita, nonché dallo sviluppo di un rilascio quantico ben definito di ACh. L’applicazione cronica di BDNF e l’aumento del cAMP intracellulare hanno provocato l’inibizione del processo di maturazione, mentre NT-3, NT-4, GDNF, HB-GAM e HGF sembrano promuoverlo. La risposta al BDNF è stata mediata dal recettore TrkB. Gli inibitori della PKA potrebbero invertire gli effetti inibitori del cAMP e del BDNF, il che suggerisce che il cAMP e il BDNF sopprimono la maturazione della NMJ attraverso la via di segnalazione cAMP-PKA-dipendente. Ulteriori indagini hanno rivelato che le CaMK e il Ca2+ intracellulare mediano gli effetti inibitori del cAMP e del BDNF.
