A existência de bactérias de biofilme em superfícies naturais e artificiais está a tornar-se cada vez mais evidente. Os biofilmes são grupos complexos de microrganismos, capazes de se adaptar e sobreviver num novo ambiente, desde que haja disponibilidade de nutrientes, tempo de incubação adequado e um meio apropriado. As estirpes bacterianas de Escherichia coli e Pseudomonas aeruginosa foram utilizadas para desenvolver biofilmes no laboratório. Estas estirpes bacterianas foram observadas utilizando a coloração de Gram, o teste da catalase e o teste da oxidase. Estas estirpes bacterianas foram enumeradas em termos de unidades de colónias viáveis às 0, 2, 4, 6 e 8 horas e incubadas a 30°C e 37°C, respetivamente. As estirpes bacterianas que cresceram até à fase estacionária foram inoculadas em placas de microtítulo de 96 poços a 0, 2, 4, 6 e 8 horas, incubadas a 30°C e 37°C, respetivamente, coradas com violeta de cristal, seguidas da medição da densidade ótica a 540nm. Os resultados mostraram que a E. coli e a P. aeruginosa são anaeróbios facultativos Gram-negativos, foram obtidos biofilmes e CPU viáveis de ambas as estirpes bacterianas. A absorvância das células aderentes das estirpes bacterianas de E. coli e P. aeruginosa aumentou à medida que o tempo aumentou de 0 a 6 horas e diminuiu às 8 horas, respetivamente.
