Os sistemas de libertação oral de fármacos continuam a ser a via de administração mais popular devido à sua versatilidade, facilidade de administração e adesão dos doentes. Foram desenvolvidos vários sistemas orais de libertação controlada para melhorar a libertação de fármacos na circulação sistémica. O trânsito gastrointestinal rápido impede a libertação completa do fármaco, conduzindo a uma diminuição da sua atividade. Os sistemas de libertação de fármacos gastro-retentores são concebidos para prolongar e atrasar o tempo de esvaziamento gástrico (GET). O GET é o fator mais importante que ajuda a forma de dosagem a permanecer no estômago durante mais tempo do que a forma de dosagem convectiva. A retenção da forma de dosagem oral no trato gastrointestinal superior (TGI) provoca um tempo de contacto prolongado do fármaco com a mucosa gastrointestinal, conduzindo a uma maior biodisponibilidade. Os fármacos com meias-vidas mais curtas, os fármacos que são eliminados rapidamente e os fármacos que são absorvidos de forma incompleta pelo intestino delgado são os candidatos adequados para serem formulados em sistemas gastro-retentores.
