Os parasitas Leishmania são responsáveis por uma série de doenças humanas conhecidas como leishmaniose, classificadas pela OMS como uma das principais doenças tropicais. A quimioterapia atualmente disponível para o tratamento é cara e altamente tóxica, além de sofrer com a crescente resistência em áreas endémicas. A expressão do gene repórter surgiu agora como uma nova ferramenta promissora que facilita a triagem de medicamentos antileishmaníacos in vitro. Neste projeto, dois compostos químicos, RAPTA EA e RAPTA EAIMI, e um extrato vegetal, PL-1, foram selecionados pela sua atividade antileishmanial contra duas espécies de Leishmania: L. tropica e L. mexicana. A anfotericina B foi utilizada como controlo positivo para garantir a fiabilidade do ensaio do medicamento e servir de referência para a atividade antileishmanial. Foram utilizados parasitas que expressam constitucionalmente a proteína fluorescente GFP para a triagem de medicamentos. A infecção foi quantificada pela medição da intensidade da fluorescência utilizando um citómetro de fluxo. O PL-1 foi mais eficaz do que o RAPTA EA ou o RAPTA EAIMI na redução da percentagem de células THP-1 infectadas com qualquer um dos parasitas, uma vez que apresentou um valor IC50 mais baixo e não foi associado a toxicidade para as células THP-1 não tratadas (> 50 µg/ml).
