O complexo promotor da anáfase (APC) é a maior Cullin-RING E3 ligase que desempenha um papel crítico no controlo do ciclo celular e é conhecido como alvo de agentes anticancerígenos. O APC regula o início da separação dos cromossomas e a saída mitótica através da degradação da securina e da ciclina B, respetivamente. A APC controla este processo através da conjugação de ubiquitina com substratos como a ciclina B e a securina. O objetivo deste estudo foi avaliar o padrão de expressão das subunidades da APC em linhas celulares cancerígenas e em doentes com LMA. Além disso, foram concebidos compostos inibidores com base na estrutura 3D da APC e os seus efeitos inibitórios foram investigados nas linhas celulares HeLa e MDA-MB-435. Para o efeito, a estrutura 3D da APC foi prevista utilizando modelos de homologia e simulação de dinâmica molecular. A interação entre as subunidades da APC foi estudada utilizando métodos de acoplamento proteína-proteína. Verificou-se que as subunidades catalíticas da APC estavam sobre-expressas em linhas celulares cancerosas e em doentes com LMA. Além disso, a montagem e a ativação das subunidades da APC foram demonstradas ao nível atómico.
